Saša M. Miladinović promovierte in Analytischer Chemie an der University of Arkansas (USA) unter der Leitung von Professor Charles L. Wilkins mit Arbeiten zur hochauflösenden Massenspektrometrie synthetischer Polymere. Anschliessend absolvierte er zwei Postdoc-Aufenthalte: an der EPFL Lausanne im Labor von Professor Yury Tsybin zur hochauflösenden Top-down-Massenspektrometrie intakter Proteine und Peptide sowie an der ETH Zürich bei Professor Ruedi Aebersold zur gezielten und ungezielten quantitativen Proteomik.

Nach mehreren Jahren als Forschungswissenschaftler in der Industrie, wo er quantitative LC-MS-Bioanalytikmethoden für kleine Moleküle, Pharmazeutika und Proteine einschliesslich monoklonaler Antikörper entwickelte, kehrte er in die akademische Forschung zurück. Heute ist er assoziierter Professor für Bioanalytische Chemie an der HES-SO Valais/Wallis und leitet dort das Mass Spectrometry and Separation Sciences Laboratory (MS³L).

Auf Bachelorstufe unterrichtet er Massenspektrometrie, Trennwissenschaften, Strukturaufklärung, Grüne Analytische Chemie und Proteomik, auf Masterstufe Bioanalytik im regulierten Umfeld sowie Biomarker-Identifizierung mittels LC-MS-Proteomik. Jährlich betreut er Bachelorarbeiten.

Forschung: Mass Spectrometry and Separation Sciences Laboratory (MS³L)

Das MS³L entwickelt und nutzt hochauflösende quantitative Massenspektrometrie in Kombination mit fortgeschrittenen Trenntechniken, um analytische Fragestellungen in biologischen, lebensmittelbezogenen, umweltrelevanten und industriellen Proben zu bearbeiten. Die Gruppe verbindet robuste Instrumentierung mit KI- und Machine-Learning-gestützter Datenauswertung; die Arbeiten werden derzeit durch ein Innosuisse-Projekt unterstützt.

  • KI- und ML-gestützte Workflows für die Datenauswertung und Methodenentwicklung in der Massenspektrometrie
  • Charakterisierung und Quantifizierung von Biopharmazeutika
  • Quantitative und qualitative Proteomik und Metabolomik

An der Schnittstelle von Analytischer Chemie und Data Science entwickelt die Gruppe skalierbare und reproduzierbare Methoden, bildet die nächste Generation analytischer Wissenschaftlerinnen und Wissenschaftler aus und arbeitet mit Partnern aus Wissenschaft, Industrie und öffentlichem Sektor zusammen.

We aim to identify biomarkers of periodontitis while in an early and still revisible phase by developing a LC-MS metabolomics workflow specifically for gingival crevicular fluid and applying it to longitudinal samples. The results will lay the foundations for a future point-of-care diagnostic test.

Horizon Europe - Scaling-up nutritional proteins from alternative sources

Description du projet:

  • Demonstrate innovative processes for the extraction and/or production of proteins for application as nutritional food, starting from alternative sources. 
  • Address efficient and cost-effective downstream separation and purification processes (when applicable), to meet the targeted quality and stability for final applications.
  • Demonstrate nutritional adequacy of the targeted product(s) and their effect on food formulation(s) according to established testing procedures.
  • Address resource efficiency and circularity aspects to increase economic and socio-environmental added value. 
  • Test the safety of developed proteins and formulations through toxicological tests, in line with EU regulatory requirements and EFSA guidelines. Moreover, identify potential regulatory gaps and provide recommendations to overcome potential bottlenecks.
  • Include a task on consumer awareness and acceptance: involve end-users (including consumers) starting from the early stages to assess market acceptance of the novel proteins and incorporate insights in product development.
  • Ensure complementarities with past and ongoing R&I projects addressing similar challenges, including projects funded under Horizon 2020 / Horizon Europe and by the BBI JU / CBE JU.

Zusammenfassung:

This study presents the development of a sophisticated liquid chromatography-mass spectrometry approach leveraging hydrophilic interaction chromatography (HILIC) for the quantification of kanamycin and spectinomycin in fermentation media. The method was validated per International Council for Harmonisation guidelines, demonstrating robust linearity, precision, and accuracy. To mitigate pronounced matrix effects common to complex fermentation matrices, sample preparation was thoroughly optimized with solid-phase extraction employing MCX sorbent, thereby enhancing recovery rates and minimizing analytical interference. The validated protocol demonstrated high correlation coefficients (R > 0.998), underscoring its robustness and reliability for the accurate quantification of antibiotics in challenging bioprocess environments, providing a valuable analytical tool for bioreactor system monitoring.

Zusammenfassung:

We evaluated a method to localize disulfide bonds in bovine pancreatic ribonuclease (RNase) by applying low pH trypsin protein digestion with a bottom-up LC-ESI-QTOF-MS approach. The goal was to minimize disulfide bond scrambling during sample preparation. By using N-ethylmaleimide (NEM) for alkylation of free cysteines, we achieved 92% sequence coverage and successfully identified the native disulfide bonds between specific cysteine residues. However, scrambled disulfide bonds were also observed. Our results indicate that while this low pH digestion method helps preserve native disulfide bonds, further refinement is needed to fully prevent disulfide bond rearrangement during sample preparation.
